Research Overview

CL-AMY is a long-acting amylin analogue designed for sustained engagement of amylin receptors (AMYR1–3) and the calcitonin receptor. Native amylin (IAPP) is co-secreted with insulin from pancreatic beta cells and contributes to post-prandial glucagon suppression, gastric motility modulation, and central satiety signaling. CL-AMY's structural modifications confer albumin binding and substantially extended pharmacokinetic profiles in rodent models.

In vitro research has characterized CL-AMY's binding affinities at AMY1R, AMY2R, AMY3R, and CTR compared to native amylin, examining differences in receptor selectivity and signaling bias. Cell-based cAMP and calcium flux assays have been used to profile its downstream signaling.

CL-AMY is most frequently studied in combination with incretin receptor agonists, investigating whether simultaneous engagement of amylin and incretin receptors produces additive or synergistic effects on energy intake, body weight, and plasma glucose in diet-induced obesity models. Research has also examined its effects on hypothalamic neuropeptide circuits involved in food intake regulation.
