Research Overview

5-Amino-1MQ (5-amino-1-methylquinolinium) is a membrane-permeable small molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that catalyzes the methylation of nicotinamide (and other pyridines) using S-adenosylmethionine (SAM) as the methyl donor. NNMT is expressed at high levels in adipose tissue and liver, and its activity affects the availability of SAM and NAD+ precursors for cellular methylation reactions.

In vitro research using adipocyte cell lines (3T3-L1, primary adipocytes) has characterized 5-amino-1MQ's inhibition of NNMT enzymatic activity, its effects on cellular SAM levels, and downstream changes in gene methylation, histone methylation marks, and lipid metabolism gene expression. Metabolomics studies have used it to map the metabolic consequences of NNMT inhibition on the 1-carbon metabolism network.

Mechanistic studies have examined 5-amino-1MQ's effects on adipocyte differentiation, fatty acid synthesis, and expression of markers associated with the thermogenic (beige/brown) fat phenotype.
