5-Amino-1MQ (5-amino-1-methylquinolinium) is a membrane-permeable small molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that catalyzes the methylation of nicotinamide (and other pyridines) using S-adenosylmethionine (SAM) as the methyl donor. NNMT is expressed at high levels in adipose tissue and liver, and its activity affects the availability of SAM and NAD+ precursors for cellular methylation reactions.
In vitro research using adipocyte cell lines (3T3-L1, primary adipocytes) has characterized 5-amino-1MQ's inhibition of NNMT enzymatic activity, its effects on cellular SAM levels, and downstream changes in gene methylation, histone methylation marks, and lipid metabolism gene expression.
