DSIP (Delta Sleep-Inducing Peptide) is a nonapeptide first isolated in 1974 from rabbit cerebral venous blood perfusate during slow-wave sleep induction by low-frequency thalamic stimulation. It contains a unique internal lactam bridge between the Asp5 and Glu9 residues that is essential for its biological activity and contributes to its distinctive conformational properties.
In vitro and animal research has examined DSIP across a range of research areas beyond sleep, including stress response modulation (DSIP reduces plasma ACTH levels in some animal models), free radical scavenging activity, and neuroendocrine regulation involving LH, GH, and cortisol in rodent preparations.
DSIP has also been studied in the context of circadian biology, where it appears to modulate activity-rest rhythms in animal models independently of classic sleep neurotransmitter systems. Its mildly amphipathic nature (water-soluble but with some lipophilic character) and brain-penetrant properties make it a useful tool for central nervous system neuropeptide research.
