Research Overview

Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is a synthetic heptapeptide analogue of the ACTH(4-7) sequence, stabilized against proteolytic degradation by a C-terminal Pro-Gly-Pro extension. It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences as part of a program to identify short peptide fragments of ACTH with neuroprotective and neurotrophic activity independent of the hormone's adrenal effects.

In vitro research has characterized Semax's effects on BDNF and NGF expression in neuronal cultures, its influence on the serotonergic and dopaminergic systems in neuronal preparations, and its interactions with NO synthase and antioxidant pathways. It has been used as a tool to study neurotrophic signaling and to compare BDNF-mimetic peptide activity.

Animal model research has investigated Semax in rodent cognitive tests (Morris water maze, radial arm maze), ischemia-reperfusion models of stroke, and chronic stress paradigms. It is frequently compared to Selank as a complementary neuropeptide with a different mechanism — Semax primarily acting via BDNF/TrkB pathways, while Selank shows immunomodulatory and GABAergic activity.
