GHRP-2 (D-Ala-D-β-Nal-Ala-Trp-D-Phe-Lys-NH₂) is a second-generation synthetic GH-releasing peptide that acts as a potent agonist at the growth hormone secretagogue receptor type 1a (GHSR-1a), also known as the ghrelin receptor. It was developed as an improved analogue of the first-generation GHRP-6 with greater potency at GH secretion and reduced food-intake stimulation in animal models.
In vitro work with GHSR-1a-transfected cells has characterized GHRP-2's binding affinity, cAMP and IP3 signaling responses, and receptor internalization kinetics. It is frequently used alongside GHRP-6 and ipamorelin as a reference compound to study structure-activity relationships at the ghrelin receptor.
Animal studies have examined GHRP-2's stimulation of pulsatile GH secretion, its synergistic effects with GHRH analogues (CJC-1295, sermorelin), and its moderate stimulation of ACTH and cortisol release—a property that distinguishes it from the more selective ipamorelin. This makes GHRP-2 useful as a pharmacological tool when studying the ghrelin receptor's broader signaling effects beyond the GH axis.
