Research Overview

MT-1 (Melanotan I, afamelanotide) is a linear synthetic analogue of α-melanocyte stimulating hormone (α-MSH) designed for increased potency and metabolic stability compared to the endogenous 13-amino acid peptide. It incorporates a norleucine substitution at position 4 and a D-phenylalanine at position 7.

MT-1 acts as a non-selective agonist at melanocortin receptors (MC1R through MC5R), with primary research interest in MC1R-mediated melanogenesis. Activation of MC1R in melanocytes drives eumelanin production, the photo-protective brown/black melanin pigment.

Unlike MT-2 (Melanotan II), MT-1 is a linear (non-cyclic) peptide with no reported activity at MC3R/MC4R in relevant doses, making it a more selective research tool for studying photoprotection mechanisms. Research has also examined its anti-inflammatory properties via MC1R signaling in immune cells.
