Oxytocin (Cys-Tyr-Ile-Gln-Asn-Cys-Pro-Leu-Gly-NH₂) is a cyclic nonapeptide neurohormone synthesized in magnocellular neurons of the hypothalamic paraventricular and supraoptic nuclei. It is released both peripherally (posterior pituitary → bloodstream) and centrally (dendrites, axon terminals) and acts at the oxytocin receptor (OXTR), a Gq-coupled GPCR widely distributed in the brain, uterus, mammary gland, and cardiovascular tissue.
In vitro receptor pharmacology research has characterized oxytocin's binding kinetics and signal transduction at OXTR compared to its structural analogue vasopressin and the vasopressin receptor subtypes V1a, V1b, and V2. Receptor selectivity studies have used radiolabeled and fluorescent oxytocin analogues to map OXTR distribution in CNS tissue preparations.
Neuroscience research has employed oxytocin in hypothalamic slice preparations, primary hippocampal neuron cultures, and ex vivo brain perfusion models to study its role in GABAergic signaling modulation, neuroplasticity, and HPA axis feedback.
