Research Overview

Vasoactive Intestinal Peptide (VIP) is a 28-amino acid neuropeptide produced in enteric neurons, hypothalamic suprachiasmatic nucleus (SCN) neurons, immune cells, and various peripheral tissues. It acts as a potent agonist at VPAC1 and VPAC2 receptors (GPCRs of the secretin receptor family), with some activity at PAC1 receptors, stimulating cAMP accumulation and downstream PKA signaling.

In vitro receptor pharmacology research has characterized VIP's binding affinities and signaling properties at VPAC1, VPAC2, and PAC1 receptors, comparing it to related peptides including PACAP-27, PACAP-38, and secretin. Studies have defined the receptor selectivity determinants within VIP's primary sequence using truncated and alanine-scan analogues.

Circadian biology research has used VIP to study its role in SCN pacemaker synchronization, using hypothalamic slice preparations and bioluminescent reporter cell lines to measure effects on PER/CRY protein oscillations and intercellular coupling. GI research has examined VIP's effects on intestinal smooth muscle relaxation, secretion, and enteric nervous system modulation in ex vivo gut preparations.
